Chondroitin sulfate as an important component of cartilage tissue, tendons, and ligaments
Chondroprotective agents are a group of medicines that protect the human joints and promote their healing processes (hondros means "cartilage" in Greek).
Chondroprotective agents strengthen the musculoskeletal system, stimulate tissue regeneration, and have general anti-inflammatory and pain-relieving effects.
Chondroitin sulfate is undoubtedly one of the most effective components of chondroprotective agents.
Chondroitin sulfate is one of the most important biopolymer components of cartilage tissue, tendons, and ligaments. Chemically, chondroitin sulfate belongs to a group of compounds consisting of long, thread-like (fibrillar) polymer chains.
The basic unit of chondroitin sulfate consists of two main fragments. The first is an oxidized form of glucose - glucuronic acid. The second is a nitrogen-containing derivative of a simple sugar, galactose, which is part of the milk sugar lactose.
The high biological activity of the group of compounds to which chondroitin sulfate belongs is due to the presence of several active functional groups and at least one strongly acidic sulfonic group. This gives the chondroitin sulfate molecule a high affinity for water (hydrophilicity) and surface-active properties.
In living systems, the strongly acidic sulfonic groups are neutralized by inorganic cations or basic amino groups in proteins. As a result, chondroitin sulfate forms strong bonds with proteins, especially collagen.
Chondroitin sulfate is often used as a chondroprotective agent primarily because it increases the strength of joint cartilage under pressure and stretching and is part of synovial fluid, which lubricates the joints, ensuring smoother and more even movement.
A distinctive feature of chondroitin sulfate is its ability to retain water and nutrients within the cartilage in the form of water-filled cavities, which provide good shock absorption.
At the same time, chondroitin sulfate promotes cartilage permeability, which is a very important factor in nourishing this tissue and removing metabolic waste products from it.
Chondroitin sulfate strengthens the cartilage lining between joint surfaces and prevents it from wearing away. It improves phosphorus-calcium metabolism in cartilage tissue.
An important property of chondroitin sulfate is its ability to weaken the effects of enzymes that destroy connective tissue. The ability of cartilage components, such as shark cartilage, to produce this effect is about 100 times greater than that of bovine cartilage.
Chondroitin sulfate has a pronounced pain-relieving effect, reduces pain and increases mobility in affected joints, and promotes the regeneration of cartilage surfaces and the joint capsule.
Biochemical analyses of blood composition and intra-articular fluid, conducted in numerous scientific studies, objectively confirm the conclusions reached.
After ingestion and partial breakdown in the gastrointestinal tract, chondroitin sulfate fragments can serve as building blocks for the synthesis of many other types of connective tissue in the body.
It is important to point out that people used to get the necessary intake of chondroitin sulfate by eating animal tendons and cartilage, especially fish, whereas consumption of these products has now declined significantly or practically disappeared. For example, how many sharks or sturgeons have you eaten yourself in the past year?
For this reason, chondroitin sulfate must be supplied to the body in its finished form or as its precursors. To this end, chondroitin sulfate is currently extracted on an industrial scale from the bones and cartilage of fish and livestock.
The most valuable are the low-molecular-weight fractions of chondroitin sulfate, which are almost completely absorbed in the gastrointestinal tract and then serve as a basis for the synthesis of cartilage and connective tissue. The cartilage of sharks, rays, sturgeons, and salmon are the most physiologically active in this respect.
Extracting pure chondroitin sulfate from natural sources is a fairly difficult technical task because it involves separating two high-molecular-weight fibrillar compounds - chondroitin sulfate and collagen. This is naturally reflected in the relatively high cost of well-purified chondroitin sulfate.
It has been proven that chondroitin sulfate and glucosamine sulfate have a synergistic effect when combined, meaning they enhance each other’s effects. Consequently, the combination of chondroitin sulfate and glucosamine sulfate, supplemented with hyaluronic acid, is most effective in slowing the progression of degenerative changes in the joints and stopping cartilage breakdown, compared with the effects of either substance alone.
The recommended intake of chondroitin sulfate for people of normal body weight, around 70-80 kg, is 1200 mg/day when taken two or three times daily. Chondroitin sulfate should be taken for at least 12 weeks to achieve a noticeable clinical effect.
Unfortunately, the bioavailability—that is, the absorption of chondroitin sulfate by the body when taken with food—is low and estimated at about 13%. This is due to the size of its molecule, which makes it difficult for it to enter the bloodstream and then reach the body's joints.
This is another reason why long-term use (up to one year) of chondroitin sulfate is necessary for sports injuries and severe joint conditions.
Unfortunately, only long-term use of chondroitin sulfate can achieve greater joint mobility, reduced pain intensity, an X-ray-confirmed increase in joint space, and an increase in the overall thickness of the cartilage plate.
